Vegetalization of the Sea - Urchin Egg by Dinitrophenol and Animalization by Trypsin and Ficin

چکیده
H E R B S T (1892) made the remarkable discovery that the differentiation of the sea-urchin egg was shifted in a vegetal direction when lithium ions were present in the sea-water. The vegetalization implies a failure of the apical tuft to appear, a displacement of the skeleton-forming cells in the animal direction, a reduction of the ectodermal region, and a corresponding enlargement of the endoderm, which often leads to exogastrulation. Strong lithium action may lead to complete endodermization of the egg. Many other substances have later been found to cause a change of differentiation, either in a vegetal or animal direction, e.g. a partial or complete animalization by treatment of unfertilized eggs with SCNor 1-ions (also SO4, Br, and tartrate, Lindahl, 1936). The animal and vegetal principles are considered as two opposite, antagonistic gradients (Runnstrom, 1928 a, b) representing different types of metabolism (Lindahl, 1936). It is of particular interest to study the effect on determination of substances which are known to interfere with metabolism in a special way. This paper will deal with the action of dinitrophenol (DNP) which acts as an inhibitor of oxidative phosphorylation, and the proteolytic enzymes trypsin and ficin.

Mechanism for Increase in Intracellular Concentration of Free Calcium in Fertilized Sea Urchin Egg
Intracellular free calcium concentration in the sea urchin egg was calculated to increase from 0.1 mM in an unfertilized egg to 1 mM in a fertilized egg 10 min after fertilization, based on measurement of the dissociation constant between free calcium and sea urchin egg homogenate. The dissociation constant between free calcium (dialyzable calcium) and homogenate of sea urchin eggs was measured...
متن کاملDetermination of developmental stages of embryo in the Sea Urchin, Echinometra mathaei
Sea Urchin is one of the most useful tools in developmental biology studies because this organism has the simplest kind of developmental stages. We aimed to determine developmental stages and timetable of Echinometra mathaei embryo (the species of Persian Gulf). The spawning of E. mathaei was induced by 0.5M KCl injection (1ml) into the coelomic cavity. After fertilization, embryos were placed ...
متن کاملFurther studies of the effects of deprivation of sulfate on the early development of the sea urchin Paracentrotus lividus.
THE morphological effects of sulfate-free medium on sea urchin embryos were described in detail by Herbst (1904). Further studies were carried out by Lindahl (1936,1942). He was the first to consider metabolic aspects of the role of sulfate in the development of the sea urchin. Immers (1956, 1959, 1961a and b, 1962) studied the distribution and function of acid mucopolysaccharides in early deve...

متن کاملCalcium-triggered fusion of exocytotic granules requires proteins in only one membrane.
We studied calcium-triggered fusion of sea urchin egg secretory granules to test whether membrane bound fusion proteins are required in both fusing membranes. Using both light scattering assays and video microscopy, we found that native granules fused to granules that had been inactivated with either trypsin or N-ethylmaleimide. Granules also fused with liposomes prepared from lipids extracted ...
